Organ culture of human intestinal biopsies for dendritic cell conditioning and cell-cell interactions

Objective. Establish a new organ culture model to analyze the role of bacteria and probiotics on the regulation of intestinal immune responses (dendritic cell conditioning, migration and cell-cell interactions).

Approach. Set up organ culture of human intestinal biopsies to preserve the polarity of the tissue (meaning an apical and basolateral face). Test different probiotics to evaluate their regulatory potential.

Deliverables:

  • Month-12, Set-up of a new organ culture model using different matrices for adhesion and Analysis of cell viability in the culture ;
  • M-24, Evaluation of cytokine response after bacterial inoculation;
  • M-36, Gene-expression profile after bacterial encounter.
